Abstract

Piroksikam merupakan obat antiinflamasi nonsteroid (NSAID) yang banyak digunakan untuk mengatasi nyeri dan inflamasi, namun penggunaannya dapat menimbulkan berbagai efek samping akibat interaksi dengan target biologis dan proses metabolisme obat. Penelitian ini bertujuan untuk menganalisis prediksi komputasi Toxicity Targets dan Toxicity Metabolism dari obat piroksikam menggunakan platform ProTox. Penelitian ini merupakan penelitian deskriptif dengan pendekatan in silico. Struktur molekul piroksikam diperoleh dari basis data PubChem dalam format Canonical SMILES, kemudian dianalisis menggunakan ProTox untuk memprediksi target toksisitas dan metabolisme toksisitas. Data dianalisis secara deskriptif berdasarkan hasil prediksi dan nilai probabilitas yang dihasilkan. Hasil penelitian menunjukkan bahwa pada parameter Toxicity Metabolism, enzim CYP2C9 diprediksi aktif dengan probabilitas 0,84, sedangkan CYP1A2, CYP2C19, CYP2D6, CYP3A4, dan CYP2E1 diprediksi tidak aktif. Analisis Toxicity Targets menunjukkan bahwa Prostaglandin G/H Synthase 1 merupakan target toksisitas utama dengan nilai Average Pharmacophore Fit sebesar 46,82% dan Average Similarity Known Ligands sebesar 78,25%. Hasil tersebut menunjukkan bahwa metabolisme piroksikam didominasi oleh CYP2C9, sedangkan target toksisitas utamanya adalah Prostaglandin G/H Synthase 1. Penelitian ini memberikan gambaran awal mengenai profil toksisitas piroksikam dan dapat menjadi dasar bagi penelitian eksperimental maupun pengembangan strategi penggunaan obat yang lebih aman.
